Antibacterial properties of cyclolinopeptides from flaxseed oil and their application on beef.

Abstract

The objective of this study was to investigate the antibacterial activity and potential mechanism of cyclolinopeptides, a type of cyclic hydrophobic peptides present in flaxseed oil. In this study, 1-Mso cyclolinopeptides B and 1-Mso, 3-Mso-cyclolinopeptides F from flaxseed oil exhibited excellent antibacterial activity against Listeria monocytogenes through destroying bacterial cell membrane. Our results indicated that cyclolinopeptides are one of the antibacterial components in flaxseed oil. Also, the application of cyclolinopeptides B and 1-Mso, 3-Mso-cyclolinopeptides F in inhibiting the microbial contamination of beef was investigated as well. Thus, our study highlights the promising potential of cyclolinopeptides to serve as food additives or food preservations due to their strong antibacterial activity against Listeria monocytogenes.

Flaxseed oil contains cyclic peptides known as cyclolinopeptides (CLs), including cyclolinopeptide A (CLA), cyclolinopeptide B (CLB), cyclolinopeptide D (CLD), cyclolinopeptide E (CLE), cyclolinopeptide F (CLF), cyclolinopeptide G (CLG), cyclolinopeptide H (CLH), and cyclolinopeptide I (CLI). Cyclolinopeptides (CLs) are known as a type of cyclic hydrophobic peptides present in flaxseed oil with a potential as endogenous antioxidants. Cyclolinopeptides have been reported to have other biological activity, such as immunosuppressive activity and osteoclastogenesis inhibition. A wide variety of peptides could act as host defense peptides with antimicrobial activity and were found in a lot of life forms from microorganisms to animals. The unique structure of cyclolinopeptides and their presence in the flaxseed Oil was the basis to investigate its antibacterial activity. In the present study, cyclolinopeptides from flaxseed oil were found to be effective if used as multi-functional food additives to control the microbial contamination in food products with high fat content. Based on the data obtained, the work confirmed that the cyclolinopeptides from flaxseed oil, especially 1-Mso-CLB and 1-Mso, 3- Mso-CLF, had a significantly antibacterial effect on L. monocytogenes. 1- Mso-CLB and 1-Mso, 3-Mso-CLF exhibited high antibacterial activity against L. monocytogenes through destroying bacterial cell membrane. Furthermore, cyclolinopeptides could effectively inhibit the growth of L. monocytogenes and prolong the shelf-life of beef. In conclusion, this investigation showed that cyclolinopeptides isolated from flaxseed oil could serve as an effective intervention to control the contamination of L. monocytogenes in meat products effectively.
